This week’s JC paper demonstrates that p120 associates with microtubules both directly and indirectly via an interaction between the p120 N-terminal domain and the heavy chain of the motor protein kinesin. And these interactions are shown to regulate both p120 function and subcelluar distribution.
These findings propose a new way via which the p120 may participate in the cadherin associated physiologic processes, such as cell development, differentiation and tumor growth. In this letter, I attach two reviews I regard might be helpful to understand the association of cadherin and p120. As cadherin, an adhesion protein, has important roles in the epidermal growth, and these roles are achieved under the help of p120 and other proteins via interactions, the novel interaction reported in this paper has great implications. To identify the specific relationship between cadherin and p120 and other relevant proteins will help a lot to better understand the mechanisms of many cellular physiologies, such as cell adhesion, differentiation and tumor growth. However, in my view, it still takes time to accomplish this job as the cross-talk network of so many factors might be very complicated and conflicts still exist on the effects of p120. As more and more evidences are yielded, there is reason for us to believe that one day we will finally have a thorough understanding on this.
